The Order of the Phoenix was an organization formed specifically to fight Voldemort. The last time the Dark Lord ran rampant, many members of the Order had sacrificed their lives.
When Voldemort said he would kill an entire family, he did exactly that. The few who escaped death were not much better off—for example, the Longbottom family. Both parents had been driven mad by the Cruciatus Curse, while Neville had been raised by his grandmother.
