From the comics, he knew that Joker's Healing Factor came from Dionysium, that the liquid was actually a type of metal like mercury, and that it was tied to some ancient secret.
But what normal person would care what color it turned? If the narration said some puddle of liquid was Dionysium, then it was. Why overthink it? At most, the comic had changed colorists.
Bobo, on the other hand, remained calm. Or perhaps he knew he would be useless in a fight, so he simply played to his strengths and used his clever mind to analyze the situation.
