In the original story, several years later, when the Third Shinobi World War broke out, Kakashi faced an Iwagakure Jonin. Upon seeing the White Fang's short blade, the man mistook him for Hatake Sakumo himself.
That meant Iwagakure still did not know of Hatake Sakumo's death by that point.
From this, it could be determined that Hatake Sakumo's mission had nothing to do with Iwagakure.
Sunagakure's stance was currently ambiguous, but at the very least, it had a ceasefire agreement with Konoha, so Sunagakure could also be ruled out.
That left only Kirigakure and Kumogakure.
Then there was Yakushi Nonō's unexplained departure. The Konoha High Command would most likely arrange for her to infiltrate one of those villages, wait for the right opportunity, and use the enemy's strength to get Hatake Sakumo killed.
Kirigakure was currently ruled by the Third Mizukage. That village was exceptionally bloody and brutal, and infiltrating it would not be easy.
By that reasoning, Kumogakure was the only possibility.
