Background

Tsunade is the granddaughter of both the First and Second Hokage and a disciple of the Third Hokage, Sarutobi Hiruzen. Although she and her younger brother Nawaki no longer used the Senju surname under Tobirama's de-clanization policy, they remained the Senju Clan's most direct surviving line. Her heritage, Sannin status, and the Senju Clan's lingering influence gave her exceptional political standing in Konoha. 42 157

Following a period in which she drank and gambled heavily, Tsunade's renewed involvement with Konoha began through medical research at Konoha Hospital. Her discussions with Shita Genichi on medical ninjutsu, transplantation, and genetic compatibility drew her back into sustained research and village affairs. 40 42

Genichi's revelation that Orochimaru and Danzo had used children in Wood Release experiments enraged her, particularly because the experiments defiled Hashirama's legacy and the village for which her family had died.
