Tsunade is a Senju descendant and the elder sister of Nawaki. When intelligence suggests that Nawaki may be targeted during a mission, she brings Uchiha Yao to Uzumaki Mito and urgently seeks a way to protect her brother. Before Nawaki departs, she gives him Hashirama’s chakra-patterned jade necklace, treating it as an important family heirloom. 101 102

After Nawaki is critically wounded, Tsunade follows Orochimaru’s team from a distance, attempts to safeguard her brother, and later becomes suspicious of Orochimaru.
