Hiruko initially pursued the Perfect Development Potion because he was dissatisfied with his ordinary physique and limited potential. He eagerly volunteered as Toru's test subject; Toru secretly added a hormone tailored to Hiruko's body, causing him to collapse after drinking the potion. 10

After awakening, Hiruko's chakra had risen by 92%. Though Toru stressed that this still left him far behind the Sannin, Hiruko saw the enhancement as proof that he could escape the limitations of normal training. 11 He became intensely devoted to Toru, while continuing as one of Danzō's disciples in Root. 11 15

Hiruko developed into Root's leading biological researcher beneath Toru, specializing in directed mutation, crossbreeding, summoned experimental organisms, and later human modification.
